KO Validated Antibody

WUHAN, China, September 17, 2026 — EnkiLife Biotech today announced the release of its KO/KD validated antibody portfolio, covering more than 1,500 targets. The portfolio is intended for research applications that require evidence of antibody specificity. Each antibody is validated against CRISPR/Cas9 knockout or siRNA knockdown samples, and validation data are provided with the products.

The announcement addresses a recognized issue in life science research. A 2015 estimate placed the annual cost of irreproducible preclinical research in the United States at approximately USD 28.2 billion, with biological reagents including antibodies accounting for 36.1% of the causes of irreproducibility. A Nature survey found that 90% of respondents agreed there was a reproducibility crisis in biological experiments, with many attributing it to poor-quality antibodies. An analysis of 614 widely used antibodies in published research found that over 50% performed poorly. These findings have contributed to interest in validation methods that provide direct experimental evidence of antibody specificity.

In knockout validation, CRISPR/Cas9 disrupts both alleles of the gene encoding the target protein, creating a genetically defined negative control. In knockdown validation, siRNA or shRNA reduces target expression at the mRNA level. A specific antibody shows complete signal loss in knockout samples or reduced signal in knockdown samples, while signal remains detectable in wild-type controls. The two methods differ in mechanism and stringency. Knockout validation involves complete gene removal at the DNA level, producing a permanent negative control. Knockdown validation reduces target expression at the mRNA level, typically by 70–95%, and is transient.

The EnkiLife portfolio includes targets in autophagy, epigenetics, signaling pathways, cancer research, immunology, neuroscience, and metabolism. According to the company, validation data include Western blot images from knockout or knockdown lysates and corresponding wild-type controls. Antibodies that fail to show expected signal loss are excluded from the portfolio. The portfolio includes rabbit monoclonal and polyclonal antibodies validated for applications such as Western blotting, immunohistochemistry, and immunofluorescence.

In 2016, an international working group published a proposal in Nature Methods outlining five conceptual pillars for antibody validation, including genetic strategies such as knockout and knockdown. The release of genetically validated antibodies reflects ongoing efforts to improve reproducibility in antibody-based research. EnkiLife Biotech is an ISO 9001:2015 certified manufacturer of research reagents for cell biology and immunology, serving customers in over 100 countries and territories.
